Intended use

This ELISpot Flex kit is intended for the enumeration of cells secreting mouse IgE (allotype a) using the ELISpot assay. For research use only. Not for use in diagnostic procedures.

Serum/Plasma samples

IgE secretion can be induced by pre-activation of cells with the included anti-CD40 mAb and IL-4. Please note that stimulation with anti-CD40/IL-4 induces switching to IgE and IgG1 and results with stimulated cells may therefore not be representative of the situation in vivo.

Mouse IgE allotypes
This kit specifically detects the IgEa allotype. Mouse strains with the IgEa allotype include e.g., AKR, BALB/c, C3H/He, and CBA. Mouse strains with the IgEb allotype include e.g., C57BL/6, C57BL/10, and SJL.

IgEa

Analyte description

Immunoglobulin E (IgE) E is secreted by plasma cells as a monomer during the adaptive immune response. IgE antibodies are believed to play an important role in the immune defense against parasitic infections and in hypersensitivity type I reactions upon re-exposure to a specific allergen by binding to and cross-linking Fc receptors (FcεRI) expressed on mast cells and basophils. Allergen-specific IgE is the basis for many in vitro assays for allergy diagnosis. In mice, two allotypes exist: IgEa and IgEb. IgEa differs by 12 single-base changes from IgEb and is present in different mouse strains.
